  • Overview
  • Citation
  • hPSC-Dopaminergic Neuron Differentiation Kit(RP01017)provides a holistic solution for the highly efficient differentiation of human pluripotent stem cells (hPSC) into mature mDAN (midbrain dopaminergic neurons). This comprehensive kit comprises the hPSC-mDAP Differentiation Kit, hPSC-mDAP Maintenance Medium, and hPSC-mDAN Maturation Medium, offering a flexible selection tailored to your research needs.

With hPSC-mDAP Differentiation Kit, researchers can obtain high-purity mDAP  with >90% Lmx1a+/ Foxa2+/ En1+ expression. The hPSC-mDAN Maturation Medium transforms mDAP into mature mDAN (TH+/ Nurr1+) with specific electrophysiological activity and dopamine release function. The differentiated human mDAP and mDAN can be used in scientific research on neurodegenerative diseases, drug screening, and cell transplantation of Parkinson's disease models.

    Process

    Cell morphology during differentiation

A, B, C: cell morphology on days 11, 12 and 45 during the differentiation process of hPSC- Dopaminergic Neuron Differentiation Kit. Scale bar: 200μm.

    D, E, F: mDAP-specific marker detection (LMX1a, FOXA2, EN1, OTX2).

    Cell morphology and identification

G, H, I: Cell fluorescence illustration of mature midbrain dopaminergic neurons (mDAN, Day 40). Scale bar: 200μm.

J: Dopamine release level test results, dopamine release >2000pg/mL.

K, L: Patch-clamp test results of hPSC-derived mature midbrain dopaminergic neurons (mDAN, Day 42).
